Agricultural intensification poses a major threat to the conservation of biodiversity and associated ecosystem services. Since non-crop habitats are regarded as important refuges for farmland biodiversity, various greening measures have been proposed to halt biodiversity loss. However, the effectiveness of these measures for biodiversity conservation is still under debate. Therefore, we here compared ground-dwelling beetle (Coleoptera) assemblages of different non-crop habitats (field margins, set-aside fields sown with wildflowers, and permanent grassland fallows) and wheat fields within an intensively used agricultural landscape in western Germany. Taxonomic diversity of Carabidae, Staphylinidae and other coleopteran families and their conservation value were higher in all non-crop habitats than on wheat fields. Surprisingly, though, different types of non-crop habitats did not differ in species richness or the number of threatened species. Thus, field margins and sown wildflower fields were as effective in promoting beetle diversity as grassland fallows. However, different non-crop habitats supported different species assemblages, and several species, in particular especially large ones, were restricted to grassland fallows. These results suggest that different greening measures are effective in promoting the biodiversity of beetles, and that permanent grassland fallows are essential for nature conservation. The fact that habitat types harbored different assemblages stresses the need to combine a variety of greening measures to yield the highest benefit for biodiversity.

Nitrate reductase (NR) from the fungus Neurospora crassa is a complex homodimeric metallo-flavoenzyme, where each protomer contains three distinct domains; the catalytically active terminal molybdopterin cofactor, a central heme-containing domain, and an FAD domain which binds with the natural electron donor NADPH. Here, we demonstrate the catalytic voltammetry of variants of N. crassa NRs on a modified Au electrode with the electrochemically reduced forms of benzyl viologen (BV2+) and anthraquinone sulfonate (AQS?) acting as artificial electron donors. The biopolymer chitosan used to entrap NR on the electrode non-covalently and the enzyme film was both stable and highly active. Electrochemistry was conducted on two distinct forms; one lacking the FAD cofactor and the other lacking both the FAD and heme cofactors. While both enzymes showed catalytic nitrate reductase activity, removal of the heme cofactor resulted in a more significant effect on the rate of nitrate reduction. Electrochemical simulation was carried out to enable kinetic characterisation of both the NR:nitrate and NR:mediator reactions.  相似文献

Substantial evidence exists for the age-related decline in maximal strength and strength development. Despite the importance of knee extensor strength for physical function and mobility in the elderly, studies focusing on the underlying neuromuscular mechanisms of the quadriceps muscle weakness are limited.The aim of this study was to investigate the contributions of age-related neural and muscular changes in the quadriceps muscle to decreases in isometric maximal voluntary torque (iMVT) and explosive voluntary strength. The interpolated twitch technique and normalized surface electromyography (EMG) signal during iMVT were analyzed to assess changes in neural drive to the muscles of 15 young and 15 elderly volunteers. The maximal rate of torque development as well as rate of torque development, impulse and neuromuscular activation in the early phase of contraction were determined. Spinal excitability was estimated using the H reflex technique. Changes at the muscle level were evaluated by analyzing the contractile properties and lean mass.The age-related decrease in iMVT was accompanied by a decline in voluntary activation and normalized surface EMG amplitude. Mechanical parameters of explosive voluntary strength were reduced while the corresponding muscle activation remained primarily unchanged. The spinal excitability of the vastus medialis was not different while M wave latency was longer. Contractile properties and lean mass were reduced.In conclusion, the age-related decline in iMVT of the quadriceps muscle might be due to a reduced neural drive and changes in skeletal muscle properties. The decrease in explosive voluntary strength seemed to be more affected by muscular than by neural changes.  相似文献

Plants differ greatly in their susceptibility to insect herbivory, suggesting both local adaptation and resistance tradeoffs. We used maize (Zea mays) recombinant inbred lines to map a quantitative trait locus (QTL) for the maize leaf aphid (Rhopalosiphum maidis) susceptibility to maize Chromosome 1. Phytochemical analysis revealed that the same locus was also associated with high levels of 2-hydroxy-4,7-dimethoxy-1,4-benzoxazin-3-one glucoside (HDMBOA-Glc) and low levels of 2,4-dihydroxy-7-methoxy-1,4-benzoxazin-3-one glucoside (DIMBOA-Glc). In vitro enzyme assays with candidate genes from the region of the QTL identified three O-methyltransferases (Bx10a-c) that convert DIMBOA-Glc to HDMBOA-Glc. Variation in HDMBOA-Glc production was attributed to a natural CACTA family transposon insertion that inactivates Bx10c in maize lines with low HDMBOA-Glc accumulation. When tested with a population of 26 diverse maize inbred lines, R. maidis produced more progeny on those with high HDMBOA-Glc and low DIMBOA-Glc. Although HDMBOA-Glc was more toxic to R. maidis than DIMBOA-Glc in vitro, BX10c activity and the resulting decline of DIMBOA-Glc upon methylation to HDMBOA-Glc were associated with reduced callose deposition as an aphid defense response in vivo. Thus, a natural transposon insertion appears to mediate an ecologically relevant trade-off between the direct toxicity and defense-inducing properties of maize benzoxazinoids.  相似文献

Many existing methods for sustainable technical product design focus on environmental efficiency while lacking a framework for a holistic, sustainable design approach that includes combined social, technical, economic, and environmental aspects in the whole product life cycle, and that provides guidance on a technical product development level. This research proposes a framework for sustainable technical product design in the case of skis. We developed a ski under the Grown brand, benchmarked according to social, environmental, economic, and technical targets, following an initial sustainability assessment, and delivered the first environmental life cycle assessment (ELCA) and the first social life cycle assessment (SLCA) of skis. The framework applies a virtual development process as a combination of ELCA to calculate the environmental footprint as carbon equivalents of all materials and processes and a technical computer‐aided design (CAD) and computer‐aided engineering (CAE) simulation and virtual optimization using parameter studies for the nearly prototype‐free development of the benchmarked skis. The feedback loops between life cycle assessment (LCA) and virtual simulation led to the elimination of highly energy intensive materials, to the pioneering use of basalt fibers in skis, to the optimization of the use of natural materials using protective coatings from natural resins, and to the optimization of the production process. From an environmental perspective, a minimum 32% reduction in carbon equivalent emissions of materials in relation to other comparably performing skis has been achieved, as well as a pioneering step forward toward transparent communication of the environmental performance by the individual, comparable, and first published ski carbon footprint per volume unit.  相似文献
